UFMylation is a newly discovered protein modification. Similar to ubiquitination, the precursor form of ubiquitin-like factor UFM1 is synthesized and cleaved by UfSP1 or UfSP2 to form activated UFM1-G83, which is then activated by a specific E1-like activating enzyme (UBA5) and transferred to E2-like conjugating enzyme (UFC1). Subsequently, UFC1 and E3-like ligase (UFL1, also known as NLBP or RCAD) jointly recognize the target protein and facilitate the specific binding between UFM1 and target protein. Finally, the UFM1-target protein covalent ligand can be cleaved by UfSP1 or UfSP2, releasing UFM1 to participate in a new cycle (Fig. 1) 2. UFMylation contributes to endoplasmic reticulum (ER) stress, DNA damage response and erythroid development by regulating protein stability and interaction.
